If problem isn't in access rights to .psafe file, some additinal info may be needed: for example, trace of file operation with Process Monitor (http://technet.microsoft.com/en-us/sysinternals/bb896645.aspx). This problem with installing virus definitions for McAfee should go away when you try again at later or different time. Hint: I can save the file with "Save as..." if I didn't modify anything, but as soon as try to modify any entry, it pops up the error message. You can do this by selecting the update function in McAfee's interface.
